Recombinant PTP1B production and allostery

Produced and characterized recombinant PTP1B to support studies of allosteric interactions and protein conformational behavior.

  • Expressed PTP1B in E. coli and generated 10+ ¹⁵N-labeled samples.
  • Purified by Ni-NTA affinity chromatography and FPLC/ÄKTA; dialyzed, concentrated, aliquoted, and stored samples.
  • Checked purity and concentration with SDS-PAGE, UV-Vis/A280, BCA, and Bradford; assessed stability with CD and thermal-shift assays.
  • Collected and analyzed biomolecular NMR data, including ¹H–¹⁵N HSQC.

  • Expressed and purified ¹⁵N-labeled Bbox1 and Bbox2, E1/E2 enzymes, and ubiquitin.
  • Contributed to ubiquitination assays and NMR binding studies.
  • Performed hands-on SDS-PAGE and Western blotting during laboratory training related to ubiquitination studies.
  • Maintained sample, purification, and QC records and shared progress in research updates.

Investigated phytochemical constituents and their predicted interaction with the SARS-CoV-2 main protease, combining analytical chemistry with computational modeling.

  • Designed and conducted the study, including sample preparation and GC-MS characterization.
  • Used molecular modeling to identify four compounds with stronger predicted inhibition than Nirmatrelvir.
  • Published the work as first author in Informatics in Medicine Unlocked (2023).
